BENOIT (Biorender)<\/em><\/figcaption><\/figure>\n\n\n\n<p class=\"has-text-align-center\">Les cellules du foie sont soumises \u00e0 des stress infectieux, m\u00e9taboliques, ioniques, m\u00e9caniques, thermiques, m\u00e9dicamenteux ou environnementaux.<br>On retrouve ainsi du stress oxydant (li\u00e9 au vieillissement ou \u00e0 des maladies), du stress osmotique et ionique, la privation en oxyg\u00e8ne ou en nutriments (isch\u00e9mie, cancer), l\u2019accumulation de lipides (st\u00e9atose), la compression (fibrose, MASH), l&rsquo;hypothermie (conservation des greffons), les chimioth\u00e9rapies anticanc\u00e9reuses (m\u00e9dicament), les nanoparticules (environnement), du stress Golgien (CDGs), ect.<\/p>\n<\/div>\n\n\n\n<div class=\"wp-container-3 wp-block-group alignwide\">\n<h3 class=\"alignwide has-text-align-center has-background-color has-text-color has-background\" style=\"background-color:#63003c\"><strong>Les th\u00e9matiques<\/strong><\/h3>\n\n\n\n<div class=\"wp-block-media-text alignwide has-media-on-the-right is-stacked-on-mobile has-foreground-color has-text-color\" style=\"grid-template-columns:auto 38%\"><figure class=\"wp-block-media-text__media\"><img loading=\"lazy\" width=\"740\" height=\"510\" src=\"https:\/\/inserm1193.universite-paris-saclay.fr\/wp-content\/uploads\/2025\/01\/Sans-titre-1-1.jpg\" alt=\"\" class=\"wp-image-2071 size-full\" srcset=\"https:\/\/inserm1193.universite-paris-saclay.fr\/wp-content\/uploads\/2025\/01\/Sans-titre-1-1.jpg 740w, https:\/\/inserm1193.universite-paris-saclay.fr\/wp-content\/uploads\/2025\/01\/Sans-titre-1-1-300x207.jpg 300w, https:\/\/inserm1193.universite-paris-saclay.fr\/wp-content\/uploads\/2025\/01\/Sans-titre-1-1-18x12.jpg 18w\" sizes=\"(max-width: 740px) 100vw, 740px\" \/><\/figure><div class=\"wp-block-media-text__content\">\n<h4 class=\"has-text-color\" style=\"color:#63003c\"><strong>Le cytosquelette : un senseur de stress et m\u00e9diateur mol\u00e9culaire des r\u00e9ponses cellulaires<\/strong><\/h4>\n\n\n\n<p class=\"has-text-align-left\">Les cellules humaines poss\u00e8dent un cytosquelette, une structure dynamique et complexe, qui intervient dans la forme 3D cellulaire, la rigidit\u00e9, la polarit\u00e9, l&rsquo;adh\u00e9rence, la division, la migration mais aussi la d\u00e9tection et la r\u00e9ponse aux stresses cellulaires.<br><br>Le cytosquelette est constitu\u00e9 de quatre composants principaux, qui sont les microfilaments d&rsquo;actine, les microtubules, les filaments interm\u00e9diaires et les filaments de septine.<br>Ces filaments peuvent interagirent entre eux et sont r\u00e9gul\u00e9s par de nombreuse prot\u00e9ines, incluant des modificateurs post-traductionnels, pour assurer les multiples fonctions physiologiques mais aussi pour s\u2019adapter \u00e0 diff\u00e9rents stress cellulaires.<\/p>\n\n\n\n<p class=\"has-cyan-bluish-gray-color has-text-color\"><em>Sch\u00e9ma : B. BAILLET<\/em><\/p>\n<\/div><\/div>\n\n\n\n<h4 class=\"has-text-color\" style=\"color:#63003c\"><strong><strong>Relocalisation des septines et chimior\u00e9sistance aux taxanes<\/strong><\/strong><\/h4>\n\n\n\n<p>Les taxanes sont des d\u00e9stabilisateurs connus des microtubules utilis\u00e9s pour le traitement des cancers du sein, de l\u2019ovaire et de la prostate. Leur efficacit\u00e9 av\u00e9r\u00e9e est cependant limit\u00e9e par des risques de chimior\u00e9sistances inn\u00e9es ou acquises.<br>Nous avons r\u00e9v\u00e9l\u00e9 que lors de r\u00e9sistance aux taxanes, les filaments de septines se relocalisent des fibres de stress d\u2019actine vers les microtubules. Ce remaniement g\u00e9n\u00e9ral du cytosquelette requiert la pr\u00e9sence de l\u2019isoforme 1 de la septine 9 et est associ\u00e9 \u00e0 la polyglutamylation de la tubuline, pour pr\u00e9server la dynamique des microtubule, la prolif\u00e9ration cellulaire et donc la chimior\u00e9sistance.<br>Le r\u00f4le pr\u00e9coce de l\u2019inactivation de Cdc42 et de la d\u00e9gradation des prot\u00e9ines BORGs par le prot\u00e9asome dans ce remaniement, ouvre des perspectives pour d\u00e9velopper de nouveaux biomarqueurs de la chimior\u00e9sistance. BAILLET<\/em><\/figcaption><\/figure>\n\n\n\n<h4 class=\"has-text-color\" style=\"color:#63003c\"><strong>Kinase de stress JNK et dynamique microtubulaire<\/strong><\/h4>\n\n\n\n<p>La kinase de stress JNK est connue pour stabiliser les microtubules dans les neurones et pour r\u00e9guler le transport mol\u00e9culaire en phosphorylant des prot\u00e9ines microtubulaires, incluant des moteurs mol\u00e9culaires.<br>Nous avons montr\u00e9 que JNK r\u00e9gule aussi la dynamique microtubulaire des cellules \u00e9pith\u00e9liales, en phosphorylant CLIP-170 sur 3 sites consensus (T25, T45 et S147). CLIP-170 est une prot\u00e9ine qui se lie \u00e0 l\u2019extr\u00e9mit\u00e9 \u00ab&nbsp;plus&nbsp;\u00bb en croissance des microtubules (+TIP) et forme des points de r\u00e9tention transitoire \u00e0 l\u2019arri\u00e8re des com\u00e8tes, au niveau des d\u00e9fauts microtubulaires. Cette prot\u00e9ine a deux fonctions connues, l\u2019une dans le recrutement sur l\u2019extr\u00e9mit\u00e9 \u2018plus\u2019 des microtubules du moteur mol\u00e9culaire Dyn\u00e9ine associ\u00e9 \u00e0 ses cargaisons et l\u2019autre dans la stimulation des \u00e9v\u00e8nement de sauvetage (repolym\u00e9risation) des microtubules.<br>En condition de stress (antibiotique, osmotique ou UVC), la prot\u00e9ine CLIP-170 est phosphoryl\u00e9e par JNK et devient un meilleur facteur de sauvetage, se retrouvant plus fr\u00e9quemment mais moins longtemps au niveau des futurs sites de sauvetage. PERDIZ<\/em><\/p>\n\n\n\n<h4 class=\"has-text-color\" style=\"color:#63003c\"><strong>Les biomarqueurs circulants des CDGs<\/strong> <strong>: des glycoprot\u00e9ines poly-modifi\u00e9es dans l&rsquo;appareil de Golgi h\u00e9patocytaire<\/strong><\/h4>\n\n\n\n<p>Les d\u00e9sordres cong\u00e9nitaux de glycosylation (CDGs) sont un groupe de maladies rares caus\u00e9es par des d\u00e9fauts de glycosylation h\u00e9rit\u00e9s. Les patients pr\u00e9sentent de nombreux sympt\u00f4mes plus ou moins s\u00e9v\u00e8res qui varient d\u2019un individu \u00e0 l\u2019autre, rendant le diagnostic difficile.<br>La glycosylation consiste en l\u2019assemblage et l\u2019ajout de cha\u00eenes saccharidiques, appel\u00e9es glycanes, sur des prot\u00e9ines ou des lipides. Elle n\u00e9cessite l\u2019activit\u00e9 de nombreuses enzymes, dont beaucoup sont localis\u00e9es dans l\u2019appareil de Golgi et requi\u00e8rent le bon fonctionnement de ce compartiment et des transporteurs\/canaux associ\u00e9s.<br>Les mol\u00e9cules glycosyl\u00e9es sont fr\u00e9quemment adress\u00e9es \u00e0 la membrane plasmique des cellules ou s\u00e9cr\u00e9t\u00e9es dans le milieu extra-cellulaire. On retrouve donc de nombreuses prot\u00e9ines glycosyl\u00e9es dans les matrices extra-cellulaires, mais aussi dans le sang, l\u2019urine ou la salive.
